วัดสัมพันธวงศ์เป็นวัดโบราณมีมาแต่สมัยอยุธยา เดิมมีคูล้อมรอบวัดเชื่อมต่อจากแม่น้ำเจ้าพระยา เรียกกันว่า วัดเกาะในรัชกาลที่ 1 เมื่อ พ.ศ. 2339 ทรงพระกรุณาให้สมเด็จพระสัมพันธวงศ์เธอ เจ้าฟ้ากรมหลวงพิทักษ์มนตรี (สมเด็จพระเจ้าหลานยาเธอ) พระนามเดิม จุ้ย ต้นราชสกุลมนตรีกุล ทำการบูรณะปฏิสังขรณ์วัดเกาะใหม่หมดทั้งพระอาราม เมื่อเสร็จเรียบร้อยแล้ว โปรดให้สถาปนาขึ้นเป็นพระอารามหลวง พระราชทานนามใหม่ว่า วัดเกาะแก้วลังการาม เพื่อเป็นเกียรติประวัติแก่สมเด็จพระพี่นางเธอ กรมพระศรีสุดารักษ์ (แก้ว) พระมารดาของกรมหลวงพิทักษ์มนตรี ในรัชกาลที่ 3 พระองค์ทรงโปรดให้ช่างหลวงทำการบูรณะปฏิสังขรณ์วัดเกาะแก้วลังการามอีกครั้งหนึ่ง ครั้นถึงสมัยรัชกาลที่ 4 ทรงเปลี่ยนนามวัดใหม่ว่า วัดสัมพันธวงศาราม วรวิหาร เพื่อเป็นการเฉลิมพระเกียรติแห่งเจ้าฟ้ากรมหลวงพิทักษ์มนตรีผู้สถาปนาวัดนี้ ในรัชกาลที่ 6 พระพินิตพินัย (แจ้ง ปิยสีโล) อดีตเจ้าอาวาสได้ขอพระราชทานพระบรมราชานุญาตซ่อมพระอุโบสถ โดยว่าจ้างช่างบูรณะใหม่และเขียนฝาผนังอุโบสถเป็นเรื่องปฐมสมโพธิ์
