โรงเรียนวัดเปาโรหิตย์สร้างขึ้นเมื่อประมาณปี พ.ศ. 2470 โดยนายชม เปาโรหิตย์ ได้ก่อตั้งโรงเรียนสำหรับสอนเด็กเล็กขึ้นโดยอาศัยศาลาวัดเป็นสถานที่เรียนหนังสือ ต่อมา พ.ศ. 2475 เจ้าพระยามุขมนตรีศรีสมุหนครบาล ได้สร้างอาคารเรียนไม้
